Overview
FOXO4-DRI is a D-retro-inverso peptide designed to interfere with the FOXO4-p53 protein interaction that keeps senescent ("zombie") cells alive. Developed by Dr. Peter de Keizer at Erasmus University Medical Center in the Netherlands, it was published in Cell in 2017.
Senescent cells accumulate with age and secrete inflammatory factors (SASP) that damage surrounding tissue and accelerate aging. FOXO4-DRI works by competitively binding to p53, preventing FOXO4 from sequestering it in the nucleus, thereby triggering selective apoptosis of senescent cells while leaving healthy cells unaffected. In aged mice, it restored fitness, fur density, and kidney function.
